The rapid antigen test COVID-19 is an immunoassay kit for the rapid qualitative determination of SARS-CoV-2 infection from sputum or posterior oropharyngeal saliva samples.

SARS-CoV-2 antigens in the samples are allowed to react with the gold conjugate coupled with anti-SARS-CoV-2 monoclonal antibodies, followed by reaction with the antibodies
anti-SARS-COV-2 monoclonals immobilized in the test line. When the sample contains SARS-CoV-2 antigens, a visible line appears in the test region on the membrane. The solution continues to migrate to encounter a control reagent that binds a control conjugate, thus producing another band in the control region.

Principle of the test:

The rapid antigen COVID-19 test is an immunochromatographic membrane test that uses highly sensitive monoclonal antibodies to detect the protein
SARS-CoV-2 nucleocapsid in sputum and posterior oropharyngeal saliva.

The test strip consists of the following parts:

• sample collection pad,
• reagent buffer,
• reactive membrane
• absorbent pad.

The reagent buffer contains colloidal gold conjugated with monoclonal antibodies against the nucleocapsid protein of SARS-CoV-2;

The reactive membrane contains the secondary antibodies against the nucleocapsid protein of SARS-CoV-2.

The entire band is fixed inside a plastic device. When the sample is added to the sample probe, the dry conjugates in the reagent buffer dissolve and migrate with the sample.

If SARS-CoV-2 antigen is present in the sample, a complex formed between the anti-SARS-2 conjugate and the virus will be captured by anti-SARS-2 monoclonal antibodies
specifics present in the region of the test line (T). The absence of the T line suggests a negative result. For procedural control purposes, a red line will always appear in the control line region (C), indicating that an adequate volume of sample has been added and membrane absorbance has occurred.

Test procedure:

WARNING! The sampling and interpretation of the results must be done only by qualified medical personnel!

Please read the instructions carefully and let the test device and samples equilibrate in temperature (15℃ - 30℃) before testing.

1. Saliva and sputum specimens from the posterior oropharyngeal segment: Use soap and water mixed with alcohol to clean your hands. The best time to collect the sample: after waking up, before brushing your teeth, eating or drinking.
2. Remove the saliva collection cup, make a "kua kua" noise from the back of the throat, removing the saliva from the back of the throat, then spit the saliva (about 2 ml) into the cup. Do not allow saliva to contaminate the outer surface of the collection cup.
3. Use the provided dropper to mix the sputum sample 8-10 times, then aspirate the sputum sample and add it to the extraction tube.
